چکیده

Previous studies have reported that the expression levels of microRNA (miR)?28?3p are downregulated in prostate cancer (PCa) compared with those adjacent normal tissues. However, to best our knowledge, function and underlying mechanisms miR?28?3p PCa not been reported. The present study aimed explore role its mechanism development PCa. In study, ADP?ribosylation factor 6 (ARF6) were analyzed using reverse transcription?quantitative PCR (RT?qPCR). Cell proliferation, colony formation, apoptosis, migration invasion determined Counting Kit?8, forming, flow cytometry Transwell assays, respectively. association between ARF6 was investigated a dual luciferase reporter assay. ARF6, Rac1, Erk1/2 phosphorylated (p)?Erk1/2 protein western blotting. results revealed downregulated, whereas upregulated cell lines (LNCaP, 22Rv?1, PC?3 DU145) line RWPE?1. overexpression promoted inhibited invasion. knockdown exerted opposite results. RT?qPCR blotting indicated target gene miR?28?3p. Finally, rescue experiments demonstrated attenuated effects mimic by upregulating Rac1 p?Erk1/2 cells. conclusion, these findings may inhibit biological ehaviors cells targeting therefore represent novel therapeutic candidate for

miR-506 inhibits cell proliferation and invasion by targeting TET family in colorectal cancer

Objective(s): Ten-eleven translocation (TET) family members have been shown to be involved in the development of many tumors. However, the biological role of the TET family and its mechanism of action in colorectal carcinogenesis and progression remain poorly understood. Materials and Methods:We measured the expression levels of TET family members in colorectal cancer (CRC) specimens, in the c...

miR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...

shRNA-mediated downregulation of α-N-Acetylgalactosaminidase inhibits migration and invasion of cancer cell lines

Objective(s): Extracellular matrix (ECM) is composed of many kinds of glycoproteins containing glycosaminoglycans (GAGs) moiety. The research was conducted based on the N-Acetylgalactosamine (GalNAc) degradation of ECM components by α-N-acetylgalactosaminidase (Nagalase) which facilitates migration and invasion of cancer cells. This study aims to investigate the effects of Naga-shRNA downregula...

miR-96 promotes cell proliferation, migration and invasion by targeting PTPN9 in breast cancer

microRNAs (miRNAs) have emerged as major regulators of the initiation and progression of human cancers, including breast cancer. The aim of this study is to determine the expression pattern of miR-96 in breast cancer and to investigate its biological role during tumorigenesis. We showed that miR-96 was significantly upregulated in breast cancer. We then investigated its function and found that ...
